What Is LGD-4033 (Ligandrol)?
LGD-4033, also known as Ligandrol or VK5211, is a non-steroidal full agonist at the androgen receptor developed by Ligand Pharmaceuticals and subsequently licensed to Viking Therapeutics. Among full-agonist SARMs, it carries the most extensive human clinical dataset — having completed Phase 1 dose-escalation and Phase 2 trials with demonstrable proof-of-concept efficacy in both healthy volunteers and hip fracture patients.
A 2010 Phase 1 trial in 76 healthy men at doses of 0.1–1.0 mg/day demonstrated dose-dependent lean body mass gains (up to ~1.21 kg at 1 mg/day over 21 days) without significant changes in PSA or hepatic enzymes. Under the name VK5211, Phase 2 trials in hip fracture patients showed significant improvements in lean body mass and physical function scores — making LGD-4033 one of the very few SARMs with clinical-stage proof-of-concept in a real disease population.
Mechanism of Action
LGD-4033 binds the androgen receptor with high affinity (Ki ~1 nM) and acts as a full agonist in skeletal muscle and bone tissue. Its selectivity over prostate and reproductive tissues is substantial, though somewhat less extreme than that of RAD-140 (which has a reported anabolic-to-androgenic ratio exceeding 90:1). The resulting anabolic gene expression profile promotes protein synthesis, nitrogen retention, and lean mass accumulation — the primary endpoints of most LGD-4033 research designs.
