What Is YK-11?
YK-11 holds a uniquely complex position in SARM research: unlike all other SARMs — which are non-steroidal — YK-11 is structurally steroidal, derived from a DHT (dihydrotestosterone) scaffold. More importantly, it exerts its biological effects through two simultaneous mechanisms: selective androgen receptor partial agonism, and myostatin inhibition via upregulation of follistatin. This dual action makes it of exceptional interest in research exploring the upper biological limits of skeletal muscle hypertrophy regulation.
Dual Mechanism: AR Agonism + Myostatin Inhibition
In vitro studies in C2C12 myoblasts have demonstrated that YK-11 increases follistatin mRNA expression dose-dependently. Follistatin is a natural antagonist of myostatin (GDF-8) — the protein that limits muscle fiber size and satellite cell activation. By simultaneously activating the androgen receptor and suppressing myostatin signaling, YK-11 potentially activates two independent muscle hypertrophy pathways that most other SARMs can only address one at a time. This makes it a valuable tool compound for research into the additive or synergistic effects of AR agonism and myostatin pathway inhibition.
Research Considerations
YK-11’s steroidal structure means it shares some androgenic properties that fully non-steroidal SARMs avoid. Some research has raised questions about potential neurotoxicity at high concentrations in cell models — an important variable to control for in in vivo study designs. Appropriate dose selection and study duration are especially critical for YK-11 research protocols.
