What Is Vinpocetine?

Vinpocetine (ethyl apovincaminate) is a semi-synthetic alkaloid derived from vincamine, the principal active compound in periwinkle (Vinca minor). It is a selective phosphodiesterase type 1 (PDE1) inhibitor and cerebral vasodilator used clinically in Eastern Europe and Asia for decades as Cavinton® (cerebrolysin equivalent) for cerebrovascular disorders and memory decline. Its primary research-relevant mechanisms include PDE1 inhibition (raising cGMP and cAMP levels in neural tissue), inhibition of voltage-gated sodium channels (mild anticonvulsant and neuroprotective effect), and enhancement of cerebral glucose and oxygen utilization.

Mechanism of Action

PDE1 hydrolyzes both cGMP and cAMP in neurons and vascular smooth muscle. By inhibiting PDE1, Vinpocetine elevates cyclic nucleotide levels, promoting vasodilation via cGMP-mediated smooth muscle relaxation and enhancing neuronal energy metabolism via cAMP-PKA signaling. It also inhibits IKKβ (IκB kinase-β), reducing NF-κB-mediated inflammatory gene expression — a mechanism that has generated interest in neuroinflammation research.
