NSI-189 Phosphate: Water-Soluble Neurogenic Compound
NSI-189 Phosphate Powder is the water-soluble phosphate salt form of NSI-189 — the investigational benzylpiperazine-aminopyridine neurogenic compound developed by Neuralstem Inc. The phosphate salt has superior aqueous solubility compared to the free base form, making it the preferred format for preparing aqueous dosing solutions for oral administration in rodent studies and for water-based cell culture assays.
Both the phosphate and free base forms stimulate hippocampal neurogenesis via the same proposed mechanism — proliferation of adult neural stem cells in the subgranular zone — and produce equivalent pharmacological effects once absorbed. The phosphate form was the salt used in the clinical Phase 1 and Phase 2 human trials.
Research Applications
- Hippocampal neurogenesis: Adult neural stem cell proliferation and differentiation, hippocampal volume studies.
- MDD and cognitive impairment: CUS rodent depression models; spatial memory tasks; hippocampal-dependent learning.
- Aqueous formulations: Preferred salt for water-based vehicle preparation at standard dosing concentrations.
- Multi-compound cognitive studies: Pairs well with acute-acting compounds like Noopept and cholinergic agents like CDP Choline.
