Sunifiram (DM-235) is a piperazinone-derived AMPAkine — a positive allosteric modulator at AMPA receptors — with reported potency approximately 1,000 times greater than piracetam per unit mass in rodent cognition assays. It also activates NMDA receptors via Src kinase-mediated GluN2B subunit phosphorylation and stimulates acetylcholine release, giving it a multi-dimensional glutamatergic and cholinergic cognitive profile. Mechanism of Action

Sunifiram enhances AMPA receptor function by slowing channel desensitization, increasing the synaptic calcium influx per glutamate release event and thereby strengthening long-term potentiation (LTP). The compound also activates PKC signaling, which facilitates NMDA receptor co-agonist site activation and amplifies synaptic plasticity cascades.
