What Is NSI-189 Free Base?
NSI-189 free base is an investigational benzylpiperazine–aminopyridine compound developed by Neuralstem Inc. as a neurogenic agent. It is studied for its ability to stimulate proliferation of adult neural stem cells in the hippocampus, increasing hippocampal volume and synaptic plasticity, and has been investigated in Major Depressive Disorder (MDD) research.
Free Base vs Phosphate
The free base is the non-salt form of NSI-189. It is more lipophilic and lower in molecular weight than the phosphate salt, while producing equivalent pharmacological effects once absorbed. The phosphate salt was the form used in the clinical Phase 1 and Phase 2 trials and is preferred for aqueous formulations; the free base suits lipophilic vehicle systems.
Research Applications
- Hippocampal neurogenesis: adult neural stem-cell proliferation and hippocampal volume studies.
- MDD & cognition: chronic-stress rodent depression models and hippocampal-dependent memory tasks.
- Neuroprotection: stroke and neurodegeneration repair models.
- Multi-compound studies: pairs with Noopept and CDP Choline.
